Gene targeted replacement of exon 1 of Cntnap2 gene with an oppositely directed neo gene. In Gao et al. 2018 (PMID 29610457) Cntnap2 null mutant mice were crossed with heterozygous transgenic mice carrying the interneuron-specific Gad1-eGFP reporter to generate Cntnap2 null mutant (test animals) and wildtype (controls) mice with the reporter. In PMID 31853066 mice were inbred after colonizing with segmented filamentous bacteria (SFB) from donor mice.

Cntnap2 null mutant mice with a temporally precise reduction in E:I balance in the medial prefrontal cortex (mPFC) generated by optogenetically increasing the excitability of inhibitory parvalbumin (PV) neurons in a real-time and reversible manner. PV:Cre homozygous mice were bred with cntnap2 KO male mice for several generations to obtain PV:Cre homozygous, Cntnap2 wildtype, heterozygous and null mice. AAV5-EF1a::DIO-SSFO-eYFP was injected into the medial prefrontal cortex to selectively express the microbial stabilized step-function opsin (SSFO) in mouse parvalbumin neurons. Activation of SSFO in PV neurons by a single pulse of blue light (2 second light pulse of wavelength 473 nm) resulted in the neurons requiring lower inward-current amplitudes to generate action potentials. This indicated SSFO activation increased the excitability of the inhibitory PV neurons.
