Mice harboring the humanized A350V mutation in the IQSEC2 gene show reduced surface expression of GluA2 AMPA receptors in mouse hippocampal tissue, reduced basal synaptic transmission in the hippocampus, increased locomotion, abnormal social behavior and spatial learning deficits.

This model was created by knocking in human exon 15 from S1474 through 107 bp 3ââ¬Â² of exon 15 of the human IQSEC2 gene in place of the corresponding region of the murine IQSEC2 gene via gene targeting in embryonic stem cells.A frameshift was introduced by the insertion of a C nucleotide immediately 5ââ¬Â²of S1474. An additional 107 bp and an exogenous SV40 polyadenylation sequence was also introduced after chimeric exon 15.
